Mitochondrial Ageing: New insights ex silico

Abstract:
Mitochondria are key organelles in many cellular functions, from respiration to cell death. It is thus not surprising that mitochondrial dysfunction is associated with many human diseases, including age-related ailments such as Sarcopenia. This gives support for the important role of mitochondria in the ageing process. In particular, mitochondrial DNA have been seen to accumulate mutations with age, and the loss of mitochondrial genome integrity is correlated with the loss of mitochondrial respiration. In this presentation, I will describe my laboratory's effort in elucidating the basic mechanisms by which mtDNA mutations occur and accumulate in cells. We have used mathematical modeling (stochastic and hybrid deterministic-stochastic models) and computer simulations to determine the most parsimonious assumptions that can explain the data. Our computational research has also revealed interesting insights regarding the origin of mtDNA mutations in life, which has not been considered widely in the ageing research.
