A Novel Procedure To Estimate the Interaction Parameters between Morphine and Ketamine

Background: in a new drug interaction model, a general outcome parameter (patients well-being ) has been defined as superposition of positive and negative effects. For two drugs, the well-being can be visualized as a function of the drug concentrations by means of a 3D surface, whereby the x and y axes are the effect compartment concentrations (Ceff) of each drug and the z-axis the well-being. In order to draw this surface for a clinically relevant drug combination, e.g. morphine and ketamine, a limited set of parameters is needed: the pharmacodynamics (PD) of the drugs and the two interaction parameters b and bs, indicating kind and strength of the interactions between the positive and the negative effects. While studies on these drugs PD are available, publications containing interaction parameters are lacking in general. Aim of this investigation is to estimate the interaction parameters basing on clinical data and, thus, to obtain the whole well-being response surface for morphine and ketamine.
