A novel direct search method for the optimization of multiple-drug regimens for pain treatment

Abstract:

Clinical regimens often involve titration of several independent variables (dosing of multiple agents, duration of treatment, number of repeated treatments, etc). Each variable may affect both the therapeutic response and treatment toxicity, and may exhibit nonlinear interactions with other variables. Systematic methods are needed to provide guidance in finding the optimal treatment, i.e. combination of variables. Direct search methods (DSM) have been proposed and successfully applied to cancer treatment and analgesia. We propose an augmented DSM to improve success rate and speed of convergence to the optimal combination for multiple-variable regimens.
