Experiment design for compartmental PK model parameter estimation

Micheal J. Avram and Tom C. Krejcie [1] derived diferent three compartment models to be used in a TCI pump which calculates the infusion rate to achieve a certain drug concentration. These concentrations should deviate minimally from the desired target. The models are based on the same set of data, but difer on sampling schedule. They compared these three compartment models with a more accurate and complex model: the recirculatory model. The conclusion was to use a brief drug infusion, instead of a rapid intravenous administration, to derive the three compartment models. Now the idea is, to have a look at diferent three compartment models derived from diferent infusion strategies. Infuse over several seconds up to minutes and compare these models with the recirculatory model as a benchmark. MATLAB was used to implement these models and to get quantitative information about them. This work is the result of a bachelor thesis performed at the Automatic Control Laboratory at the ETH Zürich. It took place during the period between September 2007 and January 2008.
